We are having trouble activating PBMCs with PMA. After Activation we lose our cell pellet. We have tried lowering the PMA, using PBS instead of HBSS, changed lysing solutions. We cannot find the problem. Any syggestions would be great. Thanks MG The protocol in short is:
Dextran treatment for sedimentation
Ficoll separation
Lysing of red blood cells
Suspend pellet in HBSS
Activation with 50ng/ml PMA (dissolved in DMSO)
Activation 30 min at 37oC
Spin and remove PMA and wash with warm HBSS
