I was just wondering if there was a lower limit when using a hemocytometer? I'd assume less than 20 cells in total to be inaccurate?

There's a nice article by Nielsen et al. (Biotechnology Progress, 1991. vol. 7, pp 560-563) on this subject. In general, I prefer adjust the dilution factor so that I count between 50 and 200 cells per chamber and then do this at least in duplicate (i.e. count the chambers on both sides of the hemocytometer). For more accuracy, it is better to count more numbers of chambers, than more cells per chamber.
