i'm working with rat skeletal myoblast cell line(L6). i'm using the fluorescence probes. it should be dissolved DMSO (100% DMSO). I'm using 2.3 microliter dmso in 1ml DMEM medium. my cells are not looking happy after the incubation with the DMSO dissolved dye. they all become rounded and going to dye. can any one tell me is that because of DMSO or I'm using ringer buffer while imaging the cells. is the ringer buffer composition matters? i would be grateful if i get the reply.

Yes, DMSO can be toxic to the cells; to check this, place 2.23 microliters of fresh DMSO without the probes on some myoblasts and look for the same reaction. If you don't see it, it may be the probes or old DMSO. Also, the composition of the Ringers can also play a role. The normal composition is:
8.6 g of NaCl, 0.3 g of KCl, and 0.33 g of CaCl2 in each 1000 ml of distilled water
Sometimes, lactated Ringer' is used instead for myoblasts/myoblasts, It has a composition of:
Sodium Chloride 6 g, Sodium Lactate 3.1 g, Potassium Chloride 300 mg, Calcium Chloride • 2H2O 20 mg brought to 1L in distilled water.
