I was wondering if anyone has encountered clogging/crustiness of purification column when trying to purify your protein from SF21's, or any other cell line. I am using SF21's for expression of my his tagged protein. Once I lyse the cells and begin to flow the cleared lysate on the Ni-column, I begin to see a crustiness and ultimately clogging of my column. I have determined that this is not do to the solubility of my protein. I didn't know if anyone else has encountered this same problem, and had any solutions?????????????
