 | Typically you would not need to add more. However, there are some cell lines that do well with a higher concentration than in standard media and if the media is nearing its expiration date you might want to consider adding some more. L-Glutamine is not immensely stable in solution. A good clue to this is if the pH of the medium has risen significantly above the roughly 7.2-7.5 range. With phenol red in the medium it will look distinctly purple instead of red-orange if the pH has gone up.
