Can anyone tell me if it is essential to use glycerol as a cropreservant for THP-1s or is it ok to just use DMSO?

 | You should be able to use DMSO at roughly 5% concentration in your standard growth medium. In fact, ATCC recommends DMSO for cryopreservation of THP-1. Just be aware that if the line has been cryopreserved with glycerol in the past, you may see a drop in viability for the first round of cells frozen with DMSO. There will have been some level of selection for optimal response to glycerol in any cells which have been preserved in glycerol. The same would apply to those which had been frozen with DMSO. And, you might want to increase the number of cells per vial a bit above that used with the glycerol stocks; maybe 20% more.
