Hello,

I have been using THP-1 cells since November but now I defreezed them and I have a different behavior as before: one part of cells grow in suspension forming aggregates and the other part are differentiated adherent cells. Before I only had suspension and single cells. Could anyone tell me if these cells grow in a normal way? If I have adherent cells, could it be that I have a bacterial contamination since I also observed some small black points in the medium ?

Hi,
Normally THP-1 cells show more aggregates in first 2 sub cultures, afterwards you can see clear single cells, this is not a problem.coming to differentiated adherent cells; i never faced this problem, only if you add PMA they turn to macrophage phenotype.just try with another frozen tube and use freshly prepared medium. all the best.
