Hi,
I'm quite new to cell culture work and have only got experience working with suspension cells. I have just got a line of THP-1s that I need to differentiate and I desperately need some tips!
I am conducting a 7-day growth curve before I differentiate them. So for now they are just contained in about 10-20ml RPMI-1640 (2mM l-glutamine, 10%FBS, 1%PenStrep) seeded at 2x10^5 cells/ml. Is there any chance they will start differentiating into macrophages and become adherent at this stage? Its just that they don't appear the same as other monocyte suspension cells that I have cultured. When I examine the flask they are not in small clumps but look kind of smokey!
I want to differentiate them next. Is there a clear protocol to follow that anyone knows of?
ANY help would be very much appreciated!!!

 | Dear Elaine..I'm Henry Puerta. Bacteriologist of the CINVESTAV who is working with THP-1 cells and their differentiation. Actually, We have got some troubles about it but maybe the following paper could help you to solve your problem "Optimized THP-1 differentiation is required for the detection of
responses to weak stimuli". I hope It could be helpfull for you.
