Over the last month or so, my bone marrow derived macrophages do not seem to look right. The appear very rounded on the plate, but are not floating or dead. In this time, I tried new FBS lots and brands (and other components to our media), and harvested the cells from many different mice, yet the problem won't go away. I have been harvesting and culturing this cell type (from the same type of mouse) for over a year with no problems. Of note, we recently moved to a new building, but cannot find any problems with our incubators, tissue culture hoods, etc, and our other cells (fibroblasts, and even BMDM's from mice on other backgrounds/genotypes) are growing fine. The only thing I can think of is mycoplasma contamination, but my mentor doesn't think that is the problem, and we have gotten these cells many times from many mice and still had the same problem. Any help, advice, or thoughts would be very much appreciated. Thanks!
