Hi, All:
I have a problem and need urgently to sort it out. I’ve stained Ki67 as a proliferation marker in the liver tissue, but couldn’t immunofluorescence stained it in the nuclei of the isolated mouse hepatocytes which fixed with 0.4% paraformaldehyde or methanol-acetone (each 50% v/v). Does anyone have any ideas on what we should do? Thanks.

 | Have you used an agent, such as Tritin 100, to permabilize the cells?

 | Thank you, Trish. I did with 0.1% Triton-100 for 10min to permabilize the cells, but it didn't work.
