Hi,
Does any one know the protocol for counting cells with a haemocytometer and methylene blue? can it be done in the same way as trypan blue or do the cells have to be fixed? (im using suspension cells)

this might give you an idea, however i would not recommend it as an assay for viability in moast mammalian cells. |

 | Methylene blue with citric acid was used to lyse cells and leave their nuclei intact and stained so they could be counted on a hemacytometer. This technique was developed before they had trypsin so cells were scraped rather than released by enzymes during harvesting. It is still a good approach to count cells that are clumped. |
