| Is that correct that we prepare the culture media using volumetric flask to measure the total volume? | | |

 | Hi
Yes,For 25com flask ,you use 5-10ml of media.75cm:20-25ml
the concentration of cells counted in different way.In dependence your work ,cells ,you make a concentration of cells.
For example,i have a 25cm flask of vero. You want to tranfer in a 75cm flask with the same concentration.
You make tripsinage and afterthat ,transfer all cells to 75cm flask with 20ml of media.Concentration is the same but the volume of flask is different.

 | I always use a graduated cylinder to bring the medium to its final volume. |

 | I don't think a volumetric flask is necessary, A graduated cylinder or beaker can be fine. If you use a beaker, you may want to calibrate the volume you use with water. When's the last time the volumetric flask has been validated?;-)-david |
