Since these (WIDR) cells are cancer and immortal, they can be split indefinitely.  Obviously, you do not want to do this since the cells can mutate.  The best bet is to make a large amount of cells in the first few days and freeze down several ampoules.  This is done in the first few splits so that you minimize genetic changes. Then, use the cell line for about a month, toss and unfreeze a new ampoule for use.  Make sure you make enough cells to complete your experiment, usually 5 or six ampoules.  Keep the seed ampoule separate so you keep track of which generation the cells are.

So--  Make about 4 seed ampoules.  Then from each seed, make several ampoules for use.  You can get about 15-20 lines this way.
