My question is what problem do a user face while loading and using Recombiant CHO cell line in hollow fiber cartridge

There should not be any problem loading or using CHO lines in a hollow fiber bioreactor. Make sure that you use a blunt nose needle to load the cells. If you use a smaller needle, you can shear the cells.
