I understood that if I use RPMI-1640 with HEPES it needs to be stored out of light

but - does HEPES alone, without media needs to be in the dark as well, or is it phototoxicity safe and can be later on mixed and used with media?

HEPES (4-(2-Hydroxyethyl)piperazine-1-ethanesulfonic acid, N-(2-Hydroxyethyl)piperazine-N′-(2-ethanesulfonic acid)) is not specifically light sensitive by itself and does not need to be stroed in the dark or in amber bottles - you can but it isn't mandatory. The issue with RMPI 1640 medium with HEPES is the the presence of tryptophan and some vitamins (nicotinic acid derivatives mainly) are known to react with short wavelength blue light and UV from fluorescent light and can generate free radicals which degrade the medium. Whether this is an actual problem in day-to-day use will depend on a lot of factors (e.g. light duration, light intensity, temperature, cell line sensitivity, concentrations, pH, etc.) and may not matter in many cases.
