I am new to cell culture and am trying to set up an experiment to look at insulin sensitivity in hepatic cells that are not well defined. the dose of insulin needed to differentiate the cells is very big and the dose that i want to use to suppress gluconeogensis is very small in comparison - i wil probably use a suppression curve. how long should i incubate the cells for in the low dose insulin ?- i am adding lactate and pyruvate to the medium. any suggestions or refernces welcome.
