I have observed granulations in the bhk cells which seems to increasing over number of passages. Though cell density seems to be okay the amount of granulations is worrisome. Iwant to know reasons and solutions for the same.
After observing xs granulations the will degrade after 3-4 days of subculturing.

yes, you are correct, over number of passages sometimes leads to granulation, especially incase of BHK. Best is to revive freshly from the stock, or else increase the amount of serum in the medium (double), and when you observe granulation, be cautious about trypsinization, try to use less amount than usual, for less period of time, and use only serum (or medium with 2X serum than usual) to deactivate the trypsin.
