Hello,

we are planning to change our cell culture to non-animal serum substitutes.

We will check for cell viability and growth rate, however I am wondering If there are any additional tests I should perform
in order to compare my cell culture containing serum and without serum?

Any ideas would be highly appreciated,

If you are growing the cells to produce some secreted or sequestered molecule, you should check to make sure that the cells are still producing it at the same or an acceptable level. The cells are likely to go through a period where they do not appear very happy as they adapt to the non-animal serum substitute. Typically you would wean the cells off of the animal serum by substituting a fraction of it for the non-animal material and live with a slower growth rate while this is happening.

Agreed, and also FACS any receptors or anything you are using/measuring, as we found that we lost about 3/4 of the receptors on our cell when we started using it.

Also, keep an eye on the metabolic changes, you can simply do this by performing few functional tests specific for the cell type under study.
