I am working on endothelial cell culture for past 2 years.i am trying to isolate endothelial cells from human umbilical cord via collagenase digestion method!every time i isolate cells, for first day the cells appeared in T-flask,as soon as i changed the media (M199+10% serum+pen/strep+Amphotericin+heparin+ ECGF) more than half cells washed off. i am using fibronectin for coating flask. beside this cells do not divide,it remains alive as long as i changed the media, i am using Endothelial cell growth factor (ECGF) from sigma.i am also getting fungal infection most of the times! Can u suggest me the any other growth factor (shd be availabe in INDIA), and any staining method by which i can see cells directly on flask. Thanx
