i got the monocyte derived dendritic cells and found that surface marker CD40, CD80 and HLA-DR have no expression and a high expression of CD1a and CD86. can anyone tell me why and what is wrong with the differentiation? i seperated the monocytes from human peripheral blood and cultured the monocytes with the hGM-CSF and hIL4 seven days, then i did flow cytometry to detect the expression of surface markers such as CD40, CD80, CD1a, CD86, HLA-DR.

Do you want immature or mature dendritic cells? I never checked for CD40 and CD80, but immature dendritic cells (what you should get after treatment with GM-CSF and IL4) should be CD1a-high, CD86-low and HLA-DR-low. If the cells mature, expression of CD86 and HLA-DR should increase. So I would guess that your cells already started to mature and thatīs why you see CD86, but no HLA-DR yet.
